Erythrocyte and Leukocyte Concentration Measurement
The determination of cell concentration is conducted through the gravimetric preparation of measurement solutions from control blood, followed by precise measurement using a Coulter Counter.
The Coulter Counter operates based on electrical impedance measurement:
- An electrically conductive solution separates two electrodes.
- When a cell passes through the capillary, the electrical resistance changes.
- Each change generates an electrical pulse, which is used to count the cells.
This method allows for high-precision, automated cell counting.
Hemoglobin Concentration Measurement
Hemoglobin analysis is performed via photometric measurements at a wavelength of 540 nm, based on the Lambert-Beer law:
A = ε * c * d
- A = Measured absorption
- ε = Molar extinction coefficient of hemoglobin
- c = Substance concentration
- d = Cuvette path length
By measuring light absorption, we determine the precise hemoglobin concentration in the control blood sample.
